Genetic Polymorphisms of GSTM1 and GSTT1 and Colorectal Tumor

[Abstract]

Objective:The carcinogenesis is related to not only environmental exposure but also individual genetic diathesis such as the polymorphisms of genes.To investigate the distribution of genetic polymorphisms of biotransformation enzyme-glutathione S-transferase(GST) such as GSTM1 and GSTT1 alone and combined genotypes in colorectal cancer patients and colorectal adenomas patients by PCR,and to explore the relationship between these polymorphic genotypes and susceptibility of colorectal tumor.Methods:A hospital-based case-control study was conducted .57 cases with colorectal caner and 52 cases with colorectal adenomas and 68 cases healthy controls were involved in this study,exclude FAP and HNPCC.The frequencies of GSTM1-null and GSTT1-null genotypes in colorectal tumor and control groups were ascertained by using polymerase chain reaction (PCR) and multiple-PCR techniques.Process the data using chi-squqre test and nonparametric test of SPSS 11.5.Results:The frequency of GSTM1-null genotype in colorectal cancer patients 70.2%was significantly higher than that 51.5% of the controls(P0.05).No difference was showed in analyzing the combined effect.The frequency of GSTM1-null genotype in colorectal cancer patients in smoking was significantly higher than that of the no smoke(P0.05).No difference was showed in analyzing the combined effect.The frequency GSTM1-null genotype and GSTT1-null genotype in colon adenomas group and rectal adenomas group and controls was no difference(P>0.05).No difference was showed in analyzing the combined effect.The opperation between GSTM1 null/ GSTT1 null in colorectal adenomas group and smoking was higher risk of colorectal cancer OR 3.200(95%CI 0.666?15.381 ), but these was no difference in statistics(P>0.05).Conclusions:1. GSTM1-null is correlated with the susceptibility of colorectal cancer.GSTT1 -null is no correlated with the susceptibility of colorectal cancer.GSTM1 null/ GSTT1 null genotype is correlated with the susceptibility of colorectal cancer.2. Alone or combined of GSTM1-null and GSTT1-null is no correlated with the susceptibility of position of colorectal cancer.3. GSTM1-null, GSTT1-null and GSTM1 null/ GSTT1 null genotype in colorectal cancer patients in smoking are correlated with the susceptibility of colorectal cancer.4. Alone or combined of GSTM1-null and GSTT1-null is no correlated with the susceptibility of colorectal adenomas.5. Alone or combined of GSTM1-null and GSTT1-null is no correlated with the susceptibility of position of colorectal adenomas.6. The opperation between GSTM1 null/ GSTT1 null in colorectal adenomas group and smoking was higher risk of colorectal cancer,but these was no difference in statistics.
